測量方式
在實驗過程中,水通過液流泵在動物室和帶光學(xué)溶解氧探針的流通容器中循環(huán)。由數(shù)字控制單元(DCU)控制三通閥,軟件控制設(shè)定系統(tǒng)的循環(huán)或沖洗模式。
在循環(huán)模式下進行呼吸測量,在沖洗模式下用水浴中的含氧水沖洗動物室。循環(huán)-沖洗-循環(huán),重復(fù)運行,直到實驗結(jié)束。沖洗和循環(huán)階段的時間由用戶在軟件中選擇。
用于小樣本的mini-AQUA
對于小型魚類和昆蟲幼蟲(~1g)的水生呼吸測量,Qubit提供了mini-AQUA套裝。改良版的Q-Box Aqua帶有小型水生呼吸室(9ml)和用于更小系統(tǒng)體積的組件。還包括一個微型室(體積為1.23ml),用于測量小于0.5g的幼蟲、卵和幼魚等非常小的樣品的耗氧率VO2。在測量期間,微型室采用間歇停止流動模式,并將溶解氧探針直接放置在該室中,從而提高系統(tǒng)的靈敏度和分辨率。
